Description:
Environmental DNA (eDNA) is transforming marine biology by enabling large-scale, cost-effective biodiversity assessments that are far less resource-intensive than traditional survey methods. By analysing eDNA, scientists can gain a more comprehensive understanding of species distribution, abundance, and ecosystem health.
The current project aims to develop and deploy an autonomous eDNA sampling system that can be seamlessly integrated into the RVI’s underway seawater system. The onboard Seagoing Instrumentation Team will be supporting the project during the voyage.

1) Characterise the Quaternary evolution of Darumbal and close neighbour Woppaburra Sea Country and integrate with traditional knowledge to contribute to understanding the cultural heritage values of the SGBR.
2) Assess the Quaternary evolution and development of the coral reefs in the SGBR.
3) Study the interaction of the modern ocean currents with the reefs and seascape of the SGBR – to help validate and improve eReefs models.
4) Atmospheric measurements for the SGBR will contribute to the Reef Restoration Adaptation Project (RRAP)

Elizabeth Shadwick The primary objective is to first deploy a new set of SOTS moorings (SOFS-15 and SAZ-28) and then recover the existing SOTS moorings (SAZ-27). Each of the SOTS moorings deliver to specific aspects of the atmosphere-ocean exchanges:
• the SAZ sediment trap mooring collects samples to quantify the transfer of carbon and other nutrients to the ocean interior by sinking particles and investigate their ecological controls.
• the Southern Ocean Flux Station (SOFS) mooring measures meteorological and ocean properties important to air-sea exchanges, ocean stratification, waves, currents and biological productivity and ecosystem structure. Water samples are collected for more detailed nutrient and plankton investigations after recovery.
Ancillary work will obtain supporting information on atmospheric and oceanographic conditions using CTD casts, and underway measurements.
1. Deploy SOFS-15 meteorology/biogeochemistry mooring
2. Deploy SAZ-28 sediment trap mooring
3. Recover SAZ-27 sediment trap mooring
4. CTD sampling (2 cast to 4550m, 2 to 600m) at the SOTS site, including collecting samples for nutrients, oxygen, dissolved inorganic carbon, alkalinity, and eDNA analyses
5. Ship meteorological observations at SOFS buoys for comparisons
6. Deployment of 2 x Argo Floats ( 1 at the nominal SOTS site, and one closer to Tasmania, in water 2000m deep, on return transit).
7. Underway air and water sampling and sensor measurements, including bio-optics and bioacoustics
8. CPR and Triaxus

Pier van der Merwe The Collaborative Australian Postgraduate Sea Training Alliance Network (CAPSTAN) is a maritime education and training initiative of CSIRO, the University of Tasmania’s Institute for Marine and Antarctic Studies (IMAS) and the Australian and New Zealand International Scientific Drilling Consortium (ANZIC). The Program is supported by grants of sea time on RV Investigator (RVI) from the CSIRO Marine National Facility and through funding from the Australian Government’s National Collaborative Research Infrastructure Strategy (NCRIS). There are no specific scientific objectives for this voyage, however the CAPSTAN platform aims to:
• Develop and provide an effective vessel‐based tertiary education experience involving national stakeholders and post-graduate students, by pooling national tertiary teaching expertise and personnel resources.
• Develop a national curriculum to standardise teaching protocols/methods and learning outcomes in conjunction with the new data collection equipment and facilities of the CSIRO research vessel RVI, the Integrated Marine Observatory System and external stakeholders, and.
• Provide and test a multi‐disciplinary research‐based teaching module for marine science postgraduates with opportunities for student mobility and national network development.
In addition to CAPSTAN’s core training objectives, this multidisciplinary voyage will explore:
1. Repeat sampling and survey of selected sites in the Bremer Canyon Region that were examined during the first Capstan voyage in 2017.
2. Oceanographic properties offshore of a persistent harmful algal bloom (HAB) in SA coastal waters.
3. Underwater Cultural Heritage (UCH) Surveys
4. Collection of samples for the AusCPR program using the Continuous Plankton Recorder.

Professor Nicole Jones Scientific objectives:
1. Develop and test novel techniques to derive, with quantified uncertainty, oceanic submesoscale currents (0.1-10 km) from remotely sensed sea surface height (SSH) and/or sea surface temperature (SST) observations.
2. Optimise data assimilation techniques for improved ocean reanalysis and forecasts.
3. Investigate lateral and vertical transport processes associated with oceanic submesoscale currents and their relation to turbulent mixing within the surface mixing layer.
4. Quantify the fluxes of nutrients to Ningaloo Reef.
5. Characterise the submesoscale current fields on the Ningaloo Coast and quantify spatial variability of eddy properties including hotspots of submesoscale activity that control ocean transport.

Operations will include:
- Deploying 3 through-water-column moorings, a wave buoy and a bottom lander frame with an acoustic release
- Deploying and recovering of WireWalker
- Surveys with shipboard ADCP and Triaxus to survey ocean features detected in HF Radar and satellite products, fixed-wing flights, drone mapping and/or satellite (SWOT and Sentinel-2) over passes
- Release of drogued drifters
- Recovering at least 2 of the through-water-column moorings and the bottom lander
- Bathymetry mapping

MNF The GSM team has seen the upgrade of multiple echosounders on board. As a team they have not had any formalised training on the use of those echosounders. The team has contracted Kongsberg Discovery to provide 12-hour shifts of training on all the newly upgraded and installed echosounders. That will leave 12 hours of ship time each night for other support teams to propose their own training routines that can utilise the ship for specific training they would like to take on.
Outcomes for the training include:
• Coverage of theory behind many of the settings within the echosounders, use cases and recommendations to uplift the teams understanding of the settings that can be changed to improve data collection
• Implementation of that theory on the echosounders during the transit to uplift the team's decision making and use of all the benthic echosounders on board
• Uplift in the use of KSync for synchronisation of echosounders
• Theory and practice on Seapath 380 inertial position and navigation system
The overall outcome for the training is to improve the way the GSM team use the equipment on board the RV Investigator ensuring they are world leading in the data they collect on the new upgraded echosounders.

MNF Marine National Facility (MNF) Trial and Calibration voyages are routinely included in the Research Vessel
Investigator (RVI) annual voyage schedule and are a critical opportunity for the MNF to undertake
maintenance, testing and calibration of vessel and scientific capabilities to ensure that the RVI can service
future research voyage schedules. The voyages are also the only allotted period within the schedule year
for the MNF to conduct trials, testing and verification of new science capabilities, and the introduction and
training of new seagoing MNF staff and affiliates.
